2. Brief

This product is used for testing Enrofloxacin in honey sample qualitatively. The whole testing need 30min, easy to operate with high sensitivity.

5. Principle

The Enrofloxacin rapid test strip is based on competitive inhibition immuno-chromatographic principle. In the flow process, Enrofloxacin in the sample combined with Enrofloxacin specific colloidal gold-labeled monoclonal antibody, Inhibit the combination between antibody and Enrofloxacin-BSA conjugate on Test line of NC membrane, lead to the color change of Test line. When the sample has no Enrofloxacin residue or concentration lower than detection limit, T line is darker than C line or T line has same color with C line; when the concentration is equal to or higher than detection limit, T line is obviously lighter than C line or T line has no color. No matter whether there is Enrofloxacin residue in sample, C line will appear, it means the test is valid.


6. Materials needed but not provided.

Equipment: homogenizer, nitrogen-drying device (or hair dryer), vortex, oscillator (optional), centrifuge, measuring pipets, balance (a sensibility reciprocal of 0.01 g).

Reagents: 50ml centrifuge tube, tips, ethyl acetate, n-hexane.


7. Sample preparation

1)Sample buffer: Dilute 10X concentrated sample buffer with deionized water at 1:9 to get sample buffer. (1.5ml 10X concentrated sample buffer + 13.5ml deionized water);

2) Take 3±0.05g honey sample into 50ml centrifuge tube, add 3ml deionized water, then add 7ml ethyl acetate, shake by hand or oscillator for 2min, centrifuge at 4000r/min for 5min;

3) Take 4ml supernatant into another centrifuge tube, blow to dry at 56℃by Nitrogen or air (or use hair dryer);

4) Add 0.5ml n-hexane, shake for 30s (or blow and beat) to dissolve the dry residues completely, then add 0.45ml sample buffer, shake by hand for 10s, be static or centrifuge at 4000r/min for 5min  until the solution is layered, discard up-layer n-hexane, absorb 200ul down-layer liquid to test.


8.Operation procedures

8.1 Read the instruction carefully before use. Return test strips and sample into room temperature.

8.2 Take bottles needed from the kit package, take out required microwells and strips, making proper marks. Please use these test strips within 1h. Seal the cap of the bottles, avoid moisture.

8.3 Take 200ul of the test samples into the micro-wells, then repeatedly absorb for many times, mix the sample with the reagent in the micro-wells completely until no solid judge by eyes (this is a very important step).

8.4 Incubate for 5min at room temperature (20-25℃), then insert the test strips into the micro-wells with the "MAX" end fully dipped in to the mixture solution.

8.5 Insert the test strips into micro-wells for 3-5min then read the result, it is invalid in other time.

9. Test Result Interpretation

9.1 Negative: T line is darker than C line, or T line has same color with C line. It means there is no Enrofloxacin residue in sample or the residue is lower than detection limit.

9.2 Positive: T line is obviously lighter than C line or T line is invisible. It means the Enrofloxacin residue is equal to or higher than detection limit.

9.3 Invalidation: C line isn’t seen wine red. It means the test card is out of efficacy, out of date or improper operation. Please run the test again using another package. If the invalid tests keep happening, please contact the supplier.    

10.Precautions

1) The test dipsticks can be used only once at room temperature, do not use test dipsticks out of expiry date.

2) Please seal the bottle after taking out required test dipsticks. If can not use all 8 micro-wells at once, cup the rest and put it back to bottle and sealed.

3) Do not touch the white membrane surface in the middle of test dipsticks, avoid sunlight and fan blowing directly.

4) Use the test dipsticks testing again for positive results.

11. Specificity

Test 500ppb Sulfonamides, Chloramphenicol and tetracyclines and other drugs. All the results are negative.


12.Storage and expiry date

Storage:  Store at 2-8 ℃ in dark, sealed, dry place, no frozen.

Expiry date: 12 months; date of production is on box.
